A horror crash that has left a teenage girl fighting for life on one of Melbourne's busiest freeways has caused peak hour traffic chaos.

There are major delays on the Monash Freeway on Tuesday morning after a single vehicle crash at Mulgrave, which left four people in hospital.

The freeway was closed almost seven hours to clear the wreckage before it reopened just after 7am.

'Delays will take some time to ease. Consider the Princes Highway as an alternative,' VicTraffic tweeted.

The delays have tested the patience of motorists.

'So we leave at 5.10am to start unloading the boats at the Melbourne Boat Show and just about to get onto the Monash freeway, only to find out that it has just been closed due to a collision. An hour and half later, we have moved about 300 metres. What a nightmare,' one posted on Facebook.

Police believe a silver Holden wagon was travelling inbound on the freeway just after midnight when it crashed into a concrete pillar.

Two teenage girls were in the back seat at the time of the crash.

One girl was taken to hospital with life threatening head injuries while the other was taken to hospital with serious injuries.

The male driver aged in his 40s was taken to hospital with non-life threatening injuries.

A woman also aged in her 40s, was taken to hospital for observation.

Firefighters had to cut three of the four occupants from the wreckage.

'They found the car had somehow found its way into the median strip between the  railings and has hit a support column on the bridge above,' Metropolitan Fire Brigade's Commander Greg Badrock told Nine News.

The freeway was closed between Wellington and Springvale Roads for almost seven hours while Major Collision Investigation Unit detectives investigated the scene.

Traffic outbound on the freeway was reduced to the two left lanes and speed was reduced to 40km/h.

The exact cause of the crash is yet to be determined.

'It is too early to say what the cause actually is before we get to speak to the occupants, they're quite seriously injured themselves,' Detective Acting Senior Sergeant Andrew Lawrence told the ABC

Anyone with information or dash cam footage of the incident is urged to contact Crime Stoppers on 1800 333 000.
