A frantic search is underway for a young female dentist who vanished from Sydney's CBD hours after the Mardi Gras parade.

Preethi Reddy, 32, was last seen on CCTV waiting in line at McDonald's in The Strand Arcade, off George Street, about 2.15am on Sunday.

Police said her failure to contact family for such a long time was out of character and they had serious concerns for her welfare.

'I just want my sister back safely, we are so worried about her,' Ms Reddy's sister Nithya told Daily Mail Australia.

'This is not like her, she would not be missing voluntarily... she's our smiling, beautiful angel.'

Police said her car, a 2016 grey Volkswagen Golf Comfortline registration DFP78P, was a crucial piece of evidence locating it could be key to finding her.

Dr Reddy was reported missing on Sunday after she didn't return home and her friends and family appealed for help on social media.

She left a dental conference about 7pm and was seen in her car an hour later at the Hotel Urban in St Leonards, her sister said.

Her family didn't know if she attended the parade, where she stayed on Saturday night, where she was on Sunday morning, or what happened to her car.

Dr Reddy talked on the phone with her family at 11.06am on Sunday and she 'gave no indication anything was wrong' and she she'd be home in a few hours.

Nithya said the last activity on her phone was detected in the Sydney CBD at 1.47pm on Sunday, according to her phone provider.

Dr Reddy grew up in Sydney and got her dental degree in 2009, and was taking courses in dental implants.

She works as a general dentist at Glenbrook Dental Surgery in the lower Blue Mountains, and was previously employed at Hurstville Dental And Physio Centre in western Sydney, and Dapto Smile Dental Centre, near Wollongong.

'Dr Reddy is known for being a highly professional yet friendly and approachable dentist who is exceptionally good with nervous patients,' her employee profile read.

'If you feel anxious about dental visits you will find Dr Reddy will go out of her way to help you relax so your dental visit is much more pleasant.' 

Anyone who saw Dr Reddy or her car after she left the conference or has any other information was urged to call the Penrith police station.
